Capsular Stain
Stains: India Ink
Methylene Blue

Bacteria: K. pneumoniae +
Staph epidermis -

Procedure:

1. Mix equal amounts of water & India Ink at one end of your slide.

2. Add a small amount of bacteria with your inoculating loop.

3. Mix the solution well with your inoculating loop.

4. Gently spread your solution to the other end of the slide using another
slide to do so.

5. Allow the smear to dry.

6. DO NOT HEAT FIX

7. After the smear is completely dry carefully add methylene blue and allow
it to sit for three minutes.

8. Rinse carefully and let the slide dry.

9. Examine under oil immersion.


Hints for success:

The suspension of bacteria; water and India Ink should be dark gray in color rather than black.

Be sure to spread the suspension until it is very thin. If you see cracks your suspension was too thick
